Design Notes
When bringing the protein to expression, keep in mind that the zinc fingers need zinc-ions (Zn2+), for when using IMAC columns like Nikkel (Ni2+). This could be a hindrance. There are protocols online which will help you bring the proteins to expression and purify them without harming the protein.
Source
The source of this part is IDT. We made the zinc-finger arrays with the help of www.zincfingertools.com. The source of the split-TEV is the iGEM Munich team 2013. the GSAT linker was obtained from part number BBa_K1159102. This part has been E.coli optimized by IDT sequence software when the purchase order was made.
